Avuncular DNA Testing: Testing an Uncle or Aunt
Published February 10, 2026 · Medically reviewed by the partner laboratory's medical director, AABB-accredited relationship-testing laboratory
An avuncular DNA test compares an alleged aunt or uncle (the alleged father's sibling) against the child. Because full siblings share about 50% of their DNA, a test between the sibling and child produces a statistical index of biological relationship — typically 90–99% conclusive, especially when the mother's sample is included.
When to use it
When the alleged father is unavailable (deceased, missing, refusing) and paternal grandparents are also unavailable, an aunt or uncle can be used.

Accuracy improvement
Including the mother's sample dramatically improves certainty by removing her contribution to the child's DNA — the test then focuses purely on the paternal side.
